Sterilize the hydrocortisone acetate by heating at 120-130C. for one hour on each of three successive days. Heat the mineral oil to 110C. and maintain the oil at this temperature for three hours. This treatment will lower the microorganism count. Suspend the hydrocortisone acetate in approximately 80% of the mineral oil and mill this suspension in a sterile ball min (using porcelain balls) until the particle size of the hormone has been reduced to 5 to 10 microns. After emptying the mineral oil suspension from the mill, tumble the ball jar and balls with one-half of the remaining mineral oil required. After one-half hour add this suspension to the main body of the hydrocortisone acetate in mineral oil. At this point in the procedure an assay should be run on the mineral oil suspension of hydrocortisone acetate. This will serve as a basis for determining whether the remaining amount of mineral oil and white petrolatum, as originally calculated, should be adjusted to correct for any decrease in the amount of |
